Depends on the bike.
On a 1000F - just go. I regularly do 700-800 Kilos a day.
On the others, a piece of closed cell foam under your *** 1/2" thick on the seat will help with pain.
Be well rested when you leave
Try to ride in off-peak traffic
Watch the weather
Wear all your gear.
Take along bug spray for your visor
Take all relevant tools, and a can of tyre goo.
chain to be well coated
The rest would be gen maintenance.
If you have one, a GPS is great, esp if you're going to unfamiliar places.
Know the relevant state laws - helmets etc.
Check your tyres
Go !
Good luck !

What I do If Im doing 100 mile+ I tape some folded kitchen roll to the inside of my windshield for easy visor cleaning. I had a packet of visor wipes there before but with gloves on, theres no chance of getting them out the pack. Might not be so great in dry weather. I used them when I had to overtake a lorry when the roads were damp. Which resulted in me getting a light misting of crap that dry's grey.

A tank bag which you can lay on is amazing. My upper back hurts after a while and laying on the tank bag while on the motorway is great.

Id also work out some tap signals for you and your other half. I wear earplugs and even with the slow driving and the visor up, I still cant hear whats being said. I prefer a tapping setup, once for 'fine', twice for 'can we stop', three for 'slow down' or none for 'ive fallen off, please make a U turn asap'.

Everything else has pretty much been covered.

Default Done that trip

Did that route a few years back 2 up on a Fazer 1000, my tips are - get a bike to bike intercom, some of that route is pretty boring It really made the journey enjoyable being able to talk (unless you dont want to of course)!
Forget camping, tent and sleeping bags etc is a hell of a load to carry two up.
Get the biggest top box available, it makes a good back rest and once you have off loaded your kit where your staying you can dump your kit into it when your off the bike. Have fun and make time to ride the Picos mountains
