600 F3 no start + backfire

Yes to both questions.
If engaging/disengaging the choke makes no difference or very little difference, then they are clogged. The F3 will barely start even in hot weather without the chike9. But as soon as the bike is warm, it will flood it out.

The jet in which you are asking about shout pass cleaner or air. Purse your lips around it or use compressed air. If it doesn't pass air, it's clogged.

I really don't think your carbs are as clean as you think they are. If a bike has sat at all with any fuel in them, the carbs have to be fully disassembled to bare maetal and soaked in carb cleaner and then blown totally free with air. Then for good measure, do it again. The forums are full of people who were sure that their carbs were clean only to find out they weren't.

Not trying to be a pain, just speaking from experience.

that was it, the jets were clogged. Nothing worked to clear them. I had to hook up a small hose to them and then carb cleaner and a blow gun. Bike run flawlessly now. So the first time you told me the carbs were still dirty you were right on. Thank you
