Assuming compression is good it's most likely a lack of spark issue since the plug is getting saturated with fuel. Make sure the plug in that cylinder is actually good, and since you already replaced the wire the next place to check is the coil. If you have a good plug and wire and have no spark on only one cylinder it's most likely a bad coil. The bike uses a waste spark system where 1 coil fires 2 cylinders, so if the coil is still firing on the other cylinder everything leading up to the coil is probably working correctly.

Try swapping the positions of the two coils and see if the non-firing cylinder changes.. if it does the coil is bad.

Stick the plug wire to your buddys arm and crank the bike. You'll have your answer quick enough....
